The Establishment of Immunoradiometric Assay for Insulin

Abstract:Two monoclonal antibodies against insulin are used.One of the antibodies is immobilized to plastic tubes as the solid phase, the other is labelled with 125I as a tracer.The sensitivity of the assay is 2.8mIU/L and the recovery is 92.3％～112.6％.The intraassay and interassay CVs are below 5％ and 10％ respectively.The correlation coefficient between measured and expected values is 0.996 after serial dilutions of the samples with high concentration of insulin.The value for normal samples (n=39) is 4.7～14.3mIU/L.Compared with RIA, the correlation is well(r=0.981)。
