Immunoradiometric Assay for TSH by Use of Monoclonal Antibody Coated Tube Method

  • TSH immunoradiometric assay(IRMA) is developed by using anti TSH monoclonal antibody coated on polystyrene tubes. Different conditions for antibody coating on the tubes are tested and compared. Under the condition of coating for overnight at 2~8 ℃, the maximum binding of the coated tubes in IRMA is obtained when the concentration of the added antibody is up to 2.1 mg/L. The temperature and time during coating have different effects on the binding of the coated tubes in IRMA at different concentration of antibody. The sensitivity of TSH IRMA is 0.05 mIU/L. The recovery is 88.4%~100.0%. The intra and inter assay CV are 7.4%~10.9% and 8.7%~10.7% respectively. The value for normal samples( n =46) is 0.3 ~ 6.2 mIU/L.
