ENZYMATIC SYNTHESIS OF α—~(32)PdATP OF HIGH SPECIFIC ACTIVITY

Abstract
This report desceibed a method for enzymaticpreparation of α-~(32)P dATP of high specific activity through a series of enzymatic reactions. The method is simple and convenient, transfer and purification of intermediate products is not necessary. It reduces manipulations of large amounts of radioactivity. The product was isolated by preparation HPLC. Its radiochemical purity is higher than 98%. The α-~(32)P dATP of high specific activity prepared by enzymatic method is satisfactorily used for the labelling of DNA (probe) by nick translation and'molecular hybridization.
