Abstract:
The breast cancer cells with high glucose metabolism were screened by
18F-FDG cell uptake experiment. The gene silencing technique was used to reduce NOX 4 expression in cells. The experiment was divided into three groups: control group, the non-specific transfection group and specific transfection group. Western blotting was used to detect differences in the expression of HIF-1α and TGF-β in three groups. The invasion ability of the three groups by transwell cell invasion assay was analyzed. The results showed that MDA-MB-231 breast cancer cells had a higher rate of glycolysis than MCF-7 tumor cells(
t=10.52,
P<0.05). The expression of HIF-1α and TGF-β and the invasiveness of cells were reduced by inhibiting the level of NOX 4. This study showed that inhibition of NOX 4 expression was able to inhibit the invasion of breast cancer cell with high glucose metabolism.
