Abstract:
With concentrated sulfuric acid as the catalytic agent,
13C
6-1,2:5,6-Di-O-isopropylidene-α-
D-glucofuranose was synthesized with
D-glucose-
13C
6 and acetone.
13C labeled 1,2:5,6-Di-O-isopropylidene-α-
D-glucofuranose was characterized by GC and MS. The reaction conditions were optimized to be as follows: the molar ratio of
D-glucose-
13C
6∶acetone = 1∶73.5, the volume of concentrated sulfuric acid = 10 mL, the reaction temperature = 25 ℃, and the reaction time = 12 h. Under these conditions, the yield of
13C
6-1,2:5,6-Di-O-isopropylidene-α-
D-glucofuranose was 75.6% (based on
D-glucose-
13C
6), with 99.2%
13C atom abundance and 98.7% chemical purity.
