Synthesis of 13C6-1,2:5,6-Di-O-isopropylidene-α-D-glucofuranose

  • 摘要: 采用浓硫酸为催化剂,以D-葡萄糖-13C6和丙酮为原料合成了13C6-1,2:5,6-O-双异丙叉-α-D-呋喃葡萄糖,并对其进行了气相色谱(GC)、质 谱(MS)分析。对合成条件进行了优化,优化后的合成条件为:葡萄糖与丙酮的摩尔比为1∶73.5,浓硫酸用量为10 mL,反应温度25 ℃,反应时间12 h,优化条件下合成的13C6-1,2:5,6-O-双异丙叉-α-D-呋喃葡萄糖收率为75.6%,产品中13C丰度为99.2 %,纯度为98.7%。

     

    Abstract: With concentrated sulfuric acid as the catalytic agent, 13C6-1,2:5,6-Di-O-isopropylidene-α-D-glucofuranose was synthesized with D-glucose-13C6 and acetone. 13C labeled 1,2:5,6-Di-O-isopropylidene-α-D-glucofuranose was characterized by GC and MS. The reaction conditions were optimized to be as follows: the molar ratio of D-glucose-13C6∶acetone = 1∶73.5, the volume of concentrated sulfuric acid = 10 mL, the reaction temperature = 25 ℃, and the reaction time = 12 h. Under these conditions, the yield of 13C6-1,2:5,6-Di-O-isopropylidene-α-D-glucofuranose was 75.6% (based on D-glucose-13C6), with 99.2% 13C atom abundance and 98.7% chemical purity.
