血清3,5,3’-三碘甲腺原氨酸酶联免疫分析试剂盒的研制

Establishment of an Enzyme Linked Immunosorbent Assay for Triiodothyronine

  • 摘要: 以辣根过氧化物酶标记链亲合素(SA),T3抗体包被微孔,T3生物素化,四甲基联苯胺(TMB)为底物,建立了3,5,3’-三碘甲腺原氨酸(T3)生物素-亲合素酶联免疫分析(T3-BA-EL1SA)方法。结果显示,本方法分析灵敏度为0.2 μg/L,批内、批间变异系数分别为7.1%~8.3%和6.5%~10.5%,回收率为98.1%~107.2%;高浓度T3血清样品系列倍比稀释后,测定值与稀释度呈线性相关,相关系数为0.995 7。本试剂盒操作简便、快速,适用于临床检测和科研应用。

     

    Abstract: A sensitive and specific ELISA for Triiodothyronine was established by using anti-T3 antibody coated on the microtiter plate,T3 biotinlated.The labeling was horseradish peroxidase(HRP)conjugate of streptavidin.TMB-H2O2 solution was used as the substrate of HRP.The sensitivity of the assay was 0.2 μg/L.The intra- and inter-assay CVs were 7.1%-8.3% and 6.5%-10.5% respectively.The analytical recovery was 98.1%-107.2%, the coefficient correlation was 0.995 7. This method for measuring T3 is rapid,sensitive and convenient.It is suitable for clinical and research application.
