白蛋白酶联免疫分析试剂盒的研制

Establishment of an Enzyme Linked Immunosorbent Assay for Albumin

摘要: 采用竞争性ELISA法检测人尿中白蛋白（Albumin,Alb）的含量。将Alb抗体包被96孔微孔板，Alb与辣根过氧化物酶结合形成酶标记物，建立了白蛋白酶联免疫分析(Alb-ELISA)方法。结果显示，本方法分析灵敏度为0.28 mg/L；批内、批间变异系数分别为2.63％～5.28%和2.40％～4.26％；回收率为95.0％～106.4％；高浓度Alb样品系列倍比稀释后，测定值与稀释度呈线性相关，相关系数为r=0.997 4。本试剂盒操作简便、快速，适用于临床检测和科研应用。

Abstract: A sensitive and specific ELISA for Albumin was established by using Albumin antibody coated on the microtiter plate, Albumin labeled horseradish peroxidase(HRP)．The sensitivity of the assay was 0.28 mg/L．The intra- and inter-assay CVs were 2.63％-5.28％and 2.40％-4.26％ respectively．The analytical recovery was 95.0％-106.4％, the coefficient correlation was 0.9974. This method for measuring Alb was rapid，sensitive and convenient．It is suitable for clinical detection and research application．
