15N标记L-胱氨酸

  • 摘要:15NH4Cl为起始原料,通过改进的Gabriel方法得到S-苄基-D,L-半胱氨酸,再经酶法拆分和解保护后得到15N标记L-胱氨酸。对影响各步反应收率的主要因素进行了考察,在较优的工艺条件下,以15NH4Cl计,15N标记L-胱氨酸收率为13.5%,产物经HPLC、红外、质谱和元素分析等证实,质谱分析表明未出现同位素丰度稀释现象。

     

    Abstract: By using 15NH4Cl as starting material, an improved Gabriel preparation method was used for the synthesis of S-benzyl -D,L -cysteine, which was further converted to S-benzyl-L-cysteine by optical resolution in the presence of aminoacylase. After the removal of protective group, the target compound 15N -L-cystine was obtained. The main controlling factors of the reactions were investigated. Under the optimized conditions, the total yield of L -cystine reached 13.5% based on 15NH4Cl consumed. The product was characterized by HPLC, IR,MS -EI and elemental analysis. The dilution of enrichment was not found in the process.
