荷人肺腺癌裸鼠的99Tcm-CD44-McAb放射免疫显像

Radioimmunoimaging of Application of the 99Tcm-CD44-McAb to Nude Mice Bearing Human Lung Adenocarcinoma

  • 摘要: 采用 99 Tcm 直接标记法标记抗 CD44 的单克隆抗体,并对标记抗体的特性进行鉴定;利用 99 Tcm-CD44-McAb 对荷人肺腺癌裸进行 放射免疫显像及体内分布研究。结果显示, 99 Tcm-CD44-McAb 的标记率为 92.3 %±4.1 % , 比活度为 2.9±0.5 TBq/g ,放化纯度为 96.2 %±3.1% 。放射免疫显像结果显示,肿瘤组织对 99 Tcm-CD44-McAb 有较高的摄取,通过感兴趣区 ( Regional Interest , ROI) 技术测得肿瘤部位与对侧相应组织(对照组)的放射性摄取比(T/NT)为2.29±0.56, 明显高于对照组(P<0.05)。标记抗体的小鼠体内分布结果与显像结果基本一致。以上研究结果说明, 99Tcm-CD44-McAb 用于荷人肺腺癌裸鼠的放射免疫显像能得到较理想的 T/NT , CD44是肺腺癌放射免疫显像研究值得推荐的目标抗原之一。

     

    Abstract: The direct labeling method of 99Tcm was applied to labeling monoclonal antibodies for CD44, and then the properties of the labeling antibodies were identified; as well as biodistribution concerning nude mice bearing lung adenocarcinoma was made by applying 99Tcm-CD44-McAb. The results indicated that the labeling yield of CD44-McAb with 99Tcm was 92.3 %±4.1 %, Specific activity of the labeled antibody was 2.9±0.5 TBq/g , and the radiochemical purity was 96.2 %±3.1%. The tumor tissue had high uptake of the labeled antibodies. The T/NT measured through ROI technique were 2.29±0.56. The biodistribution results of the labeled antibodies and their imaging results were basically identical. The results showed that the ideal target to non-target ratios could be obtained by applying 99 Tcm-CD44-McAb to radioimmunoimaging of nude mice bearing lung adenocarcinoma, and CD44 may be one of ideal target antigen to lung adenocarcinoma for radioimmunoimaging.
