Musclin impairs insulinstimulated glucose uptake in the differentiation of C
2C
12 myocytes. In these cells, musclin reduce
3HG uptake, but the expression level of GLuT4 is not altered. Studies of the myocytes revealed that the expression level of NFκB and IL6 is increased, compared with that of myocytes preincubated in a musclinfree medium. The results indicate that musclininduced insulin resistance can be studied in rat skeletal muscle in vitro. They suggest that musclininduced impairment of insulin action in these muscles is related to NFκB pathway.
