肌肉素干预大鼠骨骼肌葡萄糖摄取与NF-κB的相关性

  • 摘要: 采用同位素示踪法检测 3H标记葡萄糖( 3H G)摄取,半定量 RT PCR检测 NFκ B、 IL 6 mRNA的表达以研究肌肉素( musclin)干预大鼠骨骼肌葡萄糖摄取的机制。结果显示, musclin降低了胰岛素诱导大鼠分化的成肌细胞葡萄糖摄取, NFκ B、 IL 6 mRNA的表达水平升高。肌肉素降低胰岛素诱导大鼠骨骼肌葡萄糖摄取,可能与其对 NF-κ B的激活作用有关

     

    Abstract: Musclin impairs insulinstimulated glucose uptake in the differentiation of C2C12 myocytes. In these cells, musclin reduce 3HG uptake, but the expression level of GLuT4 is not altered. Studies of the myocytes revealed that the expression level of NFκB and IL6 is increased, compared with that of myocytes preincubated in a musclinfree medium. The results indicate that musclininduced insulin resistance can be studied in rat skeletal muscle in vitro. They suggest that musclininduced impairment of insulin action in these muscles is related to NFκB pathway.
