胰岛素免疫放射分析方法的建立

  • 摘要: 摘要:采用两株抗胰岛素单克隆抗体,一株包被聚苯乙烯试管作为固相抗体、另一株标记125I,建立了胰岛素测定的双抗体夹心免疫放射分析方法。其方法学灵敏度为2.8mIU/L;曲线范围5~160mIU/L;批内、批间变异系数分别小于5%、10%;回收率92.3%~112.6%;高浓度胰岛素血清样品系列倍比稀释后测定,测定值与稀释度呈线性相关,相关系数0.996;39例健康人血清样品测定值为4.7~14.3mIU/L,该方法与胰岛素放射免疫分析法的相关性良好(r=0.981)。

     

    Abstract: Abstract:Two monoclonal antibodies against insulin are used.One of the antibodies is immobilized to plastic tubes as the solid phase, the other is labelled with 125I as a tracer.The sensitivity of the assay is 2.8mIU/L and the recovery is 92.3%~112.6%.The intraassay and interassay CVs are below 5% and 10% respectively.The correlation coefficient between measured and expected values is 0.996 after serial dilutions of the samples with high concentration of insulin.The value for normal samples (n=39) is 4.7~14.3mIU/L.Compared with RIA, the correlation is well(r=0.981)。
