Synthesis of DTPA-2SN and Its Labeling With 99Tcm

  • 摘要: 合成了DTPA-2SN(二乙烯三胺二乙酰二磺胺),并对其化学结构进行了鉴定;制备了DTPA双酸酐(DTPAA),通过DTPAA与磺胺反应合成DTPA-2SN,并对其进行了99Tcm标记。采用n(K222):n( K2CO3)薄层层析法测定标记率,Sep-Pak C18小柱纯化,鉴定放化纯度。并观察其体外稳定性。结果显示DTPAA产率为85%,DTPA-2SN的产率为80%,二者经鉴定与其化学结构式相符。DTPA-2SN易于被99Tcm标记,标记率可达90%±3%。连续观察4h 99Tcm-DTPA-2SN放化纯度均大于90%,稳定性良好。

     

    Abstract: Diethylenetriaminepentaacetic acid bianhydride (DTPAA) is prepared at first, and then DTPA-2SN is synthesized by the incorporation of sulfanilamide (SN) into DTPA,and labeled with 99Tcm. The analysis results of DTPAA and DTPA-2SN are in accord with their molecular structure. DTPA-2SN is easy labeled with 99Tcm, the highest labeling rate of product reach up 90%±3%.and its radiochemical purity is >95 after purified by Sep-Pak C18 cartridge. 99Tcm-DTPA-2SN shows high stability in vitro, its radiochemical purities is more than 90% at 4 h in room temperature. It indicates that 99Tcm-DTPA-2SN can be used for animal experiment.
