固相萃取柱上水解法合成~(18)F-FDG

Basic Hydrolysis of 1,3,4,6-tetra-O-acetyl-2-~(18)F fluoro-D-glucose on Solid Phase Extraction

  • 摘要: 研究了一种新的固相萃取柱水解18F—FDG中间体1,3,4,6-四-O-乙酰基-2-18F-β-D-葡萄糖(18F—FDG—OAc4)的方法。用活化过的C-18萃取柱捕获中间体18F—FDG—Ac4,向柱中加入1 mL2 mol/L的NaOH,室温下2 min时99%以上的中间体可转化为18F—FDG,加入磷酸缓冲液,再经C-18柱和三氧化铝柱纯化得到供注射用18F—FDG。

     

    Abstract: A new base hydrolysis method are used for 1,3,4, 6-tetra-O-acetyl-2-18Ffluoro-D-glu-cose on solid phase extraction. The labeled intermediate is trapped on an active C-18 solid phase extraction cartridge, and hydrolyzed in cartridge with 1 mL 2 mol/L NaOH at room temperature. The results show that there are over 99% of the labeled intermediate being turned into 18F-FDG within 2 rain. It is easy to get 18F-FDG after neutralized with phosphate buffer, purified by C-18 and Alumina cartridge. The basic hydrolysis on solid extraction is a simple method for preparation of 18F-FDG.
