β射线体外诱导单层贴壁细胞凋亡生物模型的建立与吸收剂量的估算

摘要: 选用纯β-发射体32P胶体掺入人肺腺癌A549细胞体外培养体系,定时洗去干预因子,动态观察细胞凋亡,建立β射线体外诱导单层贴壁细胞凋亡的生物模型。计算单层细胞厚度,根据β面源吸收剂量估算公式估算生物模型吸收剂量。结果表明,模型稳定、重复性好,为研究核素内照射治疗实体瘤的可能机制提供了符合临床实际情况的体外观察β射线诱导细胞凋亡和吸收剂量估算的方法。Abstract: The biological model of monolayer anchorage-dependent cell apoptosis induced by in vitro β-ray irradiation is established to dynamically observe the cell apoptosis. The A549 cells of adenopulmonary carcinoma cultured in vitro are irradiated byβ emitting 32P-colloid and the interven-tional factors are rinsed out at regular intervals. The thickness of the absorbed dose of monolayer anchorage-dependent cells is measured and absorbed dose of the cells is calculated by the formula of internal dosimetry ofβ emitting 32P-colloid. The results suggest that the model has the characteristics of stability and repeatability. It provides an approach for observing the cell apoptosis induced by in vitro β-irradiation and calculating the absorbed dose of cells in clinical. And the mechanisms of internal irradiation therapy for solid tumors might be obtained.
