碘标记表皮生长因子的代谢特性

摘要: 用125I、131I标记表皮生长因子(EGF),并采用SephadexG25柱对标记物进行纯化,标记率达75.4%,放化纯度达95.2%。用昆明种小鼠作125I-EGF体内分布实验及药代动力学试验;用新西兰大白兔做131I-EGF体外显像研究。结果显示,小鼠体内注入125I-EGF后1h,肾肝组织中的药物浓度很快下降;肺组织摄取一定量的125I-EGF且保留时间长达3h;脾脏的放射性在15min达到高峰后开始缓慢下降;脑组织放射性明显低于血本底。125I-EGF的体内分布、代谢消除过程等药代动力学符合二室模型,分布半衰期T1/2α为0.3min,消除半衰期T1/2β为80.8min。新西兰大白兔体外131I-EGF显像清晰。碘标记EGF的体内、外稳定性较好。Abstract: EGF is labeled with 125I or 131I, the labeling yield is 75.4% and the radiochemical purity is >95%. The animal experiments indicate that 125IEGF distribute mainly in liver, the kidney and spleen, and clear by kidney; the concentrition of brain is lower than that of blood. It is described with a biexponential model, T1/2α and T1/2β are 0.3 and 80.8 min respectively. Their stability is very good.
