脂质体介导的~(99)Tc~m标记c-mycmRNA反义寡核苷酸的初步研究

摘要: 在 99Tcm 标记 DNA的基础上, 采用阳离子脂质体对其进行包裹, 获得了脂质体介导的 99Tcm-DNA。包裹后的标记物反义寡核苷酸、正义寡核苷酸、无义寡核苷酸的放化纯度分别为 ( 96.5±3.5) %、( 95.3± 3.2 ) %、( 94.9± 4.5) %;比活度分别为 2.0± 0.5、1.0± 0.2、1.4± 0.3GBq· g-1;介导后的寡核苷酸与水、血清孵育后, 只有极小量 99Tcm脱落, 表明其稳定性较好。Abstract: Based on the labeling c myc mRNA oligonucleotide with 99 Tc m, the labeling compounds are entrapped with liposome. The results show that their radiochemical purity and specific activity are (96.5±3.5)% and 2.0±0.5 GBq·g -1 for antisense oligonucleotides,(95.3±3.2)% and 1.0±0.2 GBq·g -1 for sense oligonucleotides, (94.9±4.5)% and 1.4±0.3 GBq·g -1 for mismatched oligonucleotides,respectively. Their stability is good.
