抗肝癌米托蒽醌免疫毫微粒球的制备及鉴定

Preparation and Determination of Mitoxantrone loaded Immunonanosphere for Anti hepatoma

  • 摘要: 采用乳化高温固化法制备载米托蒽醌 (DHAQ)的牛血清白蛋白 (BSA)毫微粒球 (NP)(DHAQ- BSA- NP)后, 应用改进的 N-琥珀酰亚胺基 - 3- (2 -吡啶二硫 )丙酸酯 (SPDP)法将抗人肝癌单克隆抗体 HAb18与 DHAQ- BSA- NP偶联, 制得抗人肝癌抗体免疫毫微粒球 HAb18- DHAQ- BSA-NP。再利用 12 5I标记的兔抗小鼠免疫球蛋白 Ig G测定 HAb18- DHAQ- BSA- NP表面结合的 HAb18数量。结果显示, HAb18偶联到毫微粒球 DHAQ- BSA- NP表面, 且每克 HAb18- DHAQ- BSA- NP表面偶联 3.5mg HAb18

     

    Abstract: After prepared mitoxantrone (DHAQ) loaded bovine serum albumin(BSA) nanosphere(DHAQ BSA NP) by emulsifying high temperature solidifying method, anti human hepatoma entire antibody immunonanosphere HAb18 DHAQ BSA NP is prepared by modified N succinimidyl 3 (2 pyridyldithio) propionate(SPDP) technique. The amount of HAb18 on the surface of HAb18 DHAQ BSA NP is determined using 125 I labelled rabbit antimouse immunoglobulin G(IgG). The results show that HAb18 is bound to the surface of nanosphere DHAQ BSA NP and the average amount of HAb18 is 3.5 mg per gram of HAb18 DHAQ BSA NP.
