RADIOIMMUNOIMAGING OF ARTERIAL AND VENOUS THROMBI IN RABBIT MODEL USING 99 Tc m LABELED MONOCLONAL ANTIFIBRIN ANTIBODY

  • 摘要: 以亚氨基噻吩盐酸盐(2 I T)法修饰抗纤维蛋白的单克隆抗体( Mc Ab) S Z63,采用99 Tcm标记葡庚糖酸钠( G H)配体交换法标记。制备实验性兔股动脉和股静脉血栓模型后,经耳缘静脉注入99 Tcm  S Z63 进行 S P E C T 显像。99 Tcm 标记非特异性鼠 Ig G 作为阴性对照。显像结果表明,兔陈旧血栓显像清晰,其最佳显像时间为注射99 Tcm  S Z63 后2~4 h;99 Tcm 标记非特异性 Ig G,未见血栓显像。因此99 Tcm  S Z63 具有活体内血栓定位导向能力,显像时间短,用于血栓性疾病的放免显像有一定的可行性。

     

    Abstract: The anti fibrin McAb SZ 63 is labeled by using the method of 2 iminothiolane modification and 99 Tc m glucoheptonate(GH) transchelation. The experimental femoral arterial and venous thrombi are prepared in rabbits, then 99 Tc m SZ 63 is injected into ear edge vein and imaged by SPECT. 99 Tc m labeled murine IgG is used as a negative control. The old arterial and venous thrombi in rabbits are clearly imaged 2 to 4 h after injection of 99 Tc m SZ 63. McAb SZ 63 would be a potential agent for diagnosis and therapy of thrombotic disease.
