~(99)Tc~m-YDPC在荷瘤裸鼠体内的分布与放射免疫显像

BIODISTRIBUTION AND RADIOIMMUNOIMAGING OF 99 Tc m\|YDPC IN NUDE MICE BEARING PROSTATE CARCINOMA

  • 摘要: Schw artz 直接还原法制备99 Tcm  Y D P C,分别采用 Lindm o 法与 Scatchard 法测定标记抗体的体外免疫活性分数及亲和常数,经尾静脉注射14.4 M Bq 99 Tcm  Y D P C于荷瘤裸鼠,22 h 后进行γ照相及体内放射性分布测定,并用荷结肠癌裸鼠进行对照。研究结果显示:99 Tcm 对 Y D P C的标记率为41% ~65% ,99 Tcm  Y D P C的免疫活性分数为 581% ,对人前列腺癌细胞株 P C3 M 的亲和常数为7.71×109 L/m ol。静脉注射99 Tcm  Y D P C 于荷人前列腺癌裸鼠22 h 后,除血液与肺外, T/ N T 均大于200,明显高于荷结肠癌裸鼠组。显像结果表明,前列腺癌裸鼠肿瘤部位有明显放射性浓聚,而荷结肠癌裸鼠肿瘤部位无放射性浓聚。表明99 Tcm  Y D P C具有良好的体外免疫活性与体内定位性能,放射性核素标记的 Y D P C 有可能应用于前列腺转移癌的临床分期诊断与导向治疗。

     

    Abstract: Tc m\|YDPC is prepared by Schwartz method. In vitro immunoreactive fraction and associate constant are measured by Lindmo , s method and Scatchard method respectively. 14.4 MBq 99 Tc m\|YDPC is injected into nude mice bearing human PC\|3M prostate cancer. The labeling yield of 99 Tc m\|YDPC is up to 41%~65%. The immunoreactive fraction and associate constant of 99 Tc m\|YDPC to prostate cancer cell line PC\|3M is 58.1% and 7.71×10\+9 L/mol respectively. The radioactivity ratio of tumor over non\|tumor tissues(T/NT) is greater than 2.00 except that of tumor over blood and lung 22 h post injection and is higher than that of control group. The results indicate radioactivity concentration on tumor site in nude mice bearing prostate cancer but no radioactivity concentration on colon tumor. The investigation demonstrates that 99 Tc m\|YDPC has an excellent immunoreactivity and tumor locating property, and the radiolabeled YDPC may be useful for the diagnosis or therapy of prostate cancer.\;
