~(153)Sm 标记 DTPA-蛋白的研究

  • 摘要: 研究了双功能联接剂CADTPA与牛血清白蛋白(BSA)、链霉亲和素(SA)、CEA单抗和IgG的偶联反应,建立了153Sm标记蛋白的方法,选择了最佳标记条件,检测了其中一些偶联物的生物活性,测定了153Sm通过双功能联接剂标记蛋白的体外血清稳定性。结果表明:DTPA-SA的生物素结合活性保存率为56.5%;DTPA-CEA单抗的免疫活性与CEA多抗相近。153Sm标记CEA单抗的比活度达2.35GBq/g。153Sm标记IgG在体外血清中很稳定,41h内没有产生游离153Sm。

     

    Abstract: STUDY ON THE LABELING OF DTPAPROTEIN WITH153SmChen Lai Bao Borong(School of Chemistry and Chemical Engineering, Shanghai University,201800)Zhuang Daoling Zhou Zhentang(Shanghai Institute of Nuclear Research, 201800)Li Guiping Fong Guowei Li Biao Shi Ailan Zhu Chengmo(Ruijin Hospital,Shanghai 200025)ABSTRACTThe conjugating reaction of CADTPA with BSA、 SA、 CEAMAb and IgG is studied, the labeling method of proton with 153Sm is established,and the better conditions are selected. The biological activity of some conjugates is detected, and the serum stability of 153SmDTPAIgG is examined. Experimental results indicate the retained bioreactivity of DTPASA with biotin is 56.5%, and the immunoreactivity of DTPAMAb is the same as PAb. The MAb labeled with 153Sm has a specific activity of 2.35 GBq/g. 153SmDTPAIgG is very stable in mice serum, without releasing free 153Sm within 41 hours.Key words labeling of 153Sm CADTPA BSA CEAMAb IgG conjugating reaction bioreactivity immunoreactivity
