PREIRRADIATIONGRAFTINGOFBSA┐NASICONJUGATESONAGAROSEANDITSAPPLICATIONYiMinZhangBaolinHaHongfei(DepartmentofTechnicalPhysics,PekingUniversity,Beijing100871)ABSTRACTBSA-NASIconjugatesarepreparedat37℃for2h.ThemolarratioofBSAandNASIintheconjugatesis1∶6.PreiradiationofSepharose-4Biscariedoutatroomtemperatureinairandgraftingfolowedisperformedbelow37℃for5hunderbubblingnitrogen.Thetotaldoseisaround5kGy.ThecombinationconstantskbetweenL-,D-,D,L-tryptophansandBSAinconjugatesaremeasuredandcalculatedseparately.Theyare:kL=1.8×104L·mol-1,kD=0.0andkD,L=2.3×103L·mol-1.Thekvaluesarenotafectedbyreactiontemperature.ThegraftingcopolymerasfilingmaterialsincolumnisusedtoseparateD-,L-tryptophans.Itshowsthatinasingleseparationabout25%ofL-tryptophancouldbeseparatedfromthenatu-raltryptophans.Itindicatesthatthismethodmayapplyweltobioseparationwithfurtherim-provements.Keywordsagarosebovineserumalbumin(BSA)N-acryloxysuccinimide(NASI)tryptophanconjugationpreiradiationgraftingcolumnchromatography
