金属硫蛋白的简便提取与分离

  • 摘要: 预先给家兔注射CdCl2以诱导金属硫蛋白(Metalothionein,MT)的合成,取肝脏匀浆、离心,综合采用加热分离和酒精沉淀分离、经SephadexG-50、DEAE-Sepharose和SephadexG-25柱先后层析,即可获得高纯度的两个MT亚型。经109Cd示踪证实,该方法可靠;经HPLC分析,所得MT产品纯度达HPLC纯。该方法采用低速离心、紫外监测,无需测金属离子含量来确定MT的组分,方法简单、快速,适于一般实验室对MT的提取与分离。
