血清总雌三醇的放射免疫测定

摘要: 用雌三醇（Ｅ＿３）－６－肟－ＢＳＡ为免疫原，制备了特异性强和亲和力高的抗血清。除了与雌二醇（Ｅ＿２）及雌酮（Ｅ＿１）的交叉反应率分别为０．２％和０．１５％外，和其它８种类固醇激素的交叉反应率均＜０．００１％。亲和常数Ｋ＿ａ＝２．７×１０￣（１０）ｌ／ｍｌ。用此抗血清和￣（１２５）Ｉ－６－Ｅ＿３－肟－碘代组胺建立了血清总雌三醇直接放射免疫分析法（ＲＩＡ）并测定１４７例不同孕周正常妇女血清总Ｅ＿３含量，结果满意。

Abstract: RADIOIMMUNOASSAY OF SERUM TOTAL ESTRIOL Dai Xun Ji Di Guo Jian（Jiuxianqiao Hospitol, Beijing 100016）ABSTRACT The andsera of high specificity and fine avidity are prepared with estriol E_3-6-oxime-BSA as a im-munogen. The cross-reactivity of estradial（E_2 ） and estrone（E_1 ）with the antiserum is 0．2％ and0．15％respectively.The cross-reactivity of the other eight steroids hormones with the antisera are＜0．0001％. Avidity constant of antiserum k_a is 2．7×10~(10)l／mol．So，the direct RIA of serum total estriol is found with ~(125)I-E_3-6-oxime-iodohistamine and antiserum，The contents of total estriol are determined for the normal women with different weeks of pregnancy.
