放射配体结合分析法测定外周血白细胞GCR——探讨在白血病诊治中的意义

THE ASSAY OF GLUCOCORTICOID RECEPTOR OF PERIPHERAL LEUKOCYTES USING RADIOLIGAND BINDING ASSAY——DIAGNOSTIC VALUE IN LEUKEMIA TREATMENT AND PROGNOSIS

  • 摘要: 研究了人外周血白细胞糖皮质激素受体(GCR)检测方法的最佳条件.检测了29例正常人及80例急性白血病患者外周血白细胞GCR水平.K_d值分别为5.l±2.01nmol/l和5.38±1.21nmo/1.重点探讨了GCR与激素联合化疗疗效及近期预后的关系,结果提示凡患者在疗前GCR>6000位点/细胞,近期预后都好(16/16),凡<6000位点/细胞者,近期预后均差(14/l4).对非淋巴细胞白血病患者疗前GCR的量与疗效及预后无密切关系.报道了15例正常人及10例白血病患者外周血淋巴细胞及白细胞糖皮质激素低亲和力结合位点(GCSBL)的测定,其K_d值分别为0.8224±0.241μmol/1和1.067±0.496μmol/l分别为 3614.43±1233.33 fmol/10~7细胞和3488.94±2272.56 fmol/10~7细胞.

     

    Abstract: The method of measuring the glucocorticoid receptor (GCR) is studied by using whole peripheral leukocytes, and the GCR level of 29 normal cases and 80 cases of acute leukemia are measured. The Kd are 5. 1±2. 01 nmol/1 and 5. 38±1. 21 nmol/1 respectively. The study is emphasized in the relationship between GCR level and the effect of chemotherapy or its short-term prognosis, the result indicates that patients with GCR higher than 6000 site/cell have good short-term prognosis (16/16), whereas less than 6000 site/cell have poor short-term prognosis (14/14), and there is no such obvious relationship in the cases of non- acute leukemia. The low- affinitive binding site of glucocorticoid (GCSBL) in the peripheral lymphocytes in 15 normal cases and 10 cases of acute leukemia, the Kd are 0. 8224±0. 241 μmol/l and 1. 067±0. 496 μmol/1 respectively, and the R0 ate 3614. 43±1233. 33 fmol/107 cell and 3488. 94±2272. 56 fmol/107 cell as well.
