~(211)At标记CEA-McAb的HPLC分离分析研究

IDENTIFICATION AND SEPARATION OF ~(211)At LABELED CEA-McAb BY HYDROPHOBIC INTERACTION HPLC

  • 摘要: 利用HPLC进行了放射性核素(~(211)At,~(231)I)标记McAb的分离分析研究,采用1.0mol/l(NH_4)_2-SO_4-0.1mol/l PBS液梯度淋洗。先后经225nm紫外和放射性检测结果证明,~(211)At-CEA-M_(?)Ab偶联物与原始CEA-McAb的紫外峰一致,均在R_t=13.40±0.05min出现.而副产物及其它物质的紫外峰滞留时间与之完全不同。此外,定量地测定了McAb含量和峰面积的关系以及放射性计数。
    Abstract: The hydrophobic interaction HPLC has been used to identify the labelling procedure and to separate the main product ~(211)At-CEA-McAb. The same retention time was obtained by chromatography for CEAMcAb as well as for ~(211)At-CEA-McAb conjugate. It was demonstrated that the McAb contents and cpm of ~(211)At labeled monoclonal antibody could be measured quantitatively.
