流气式微波激活法合成氚标记化合物

A MICROWAVE ACTIVATION METHOD WITH FLOWING TRITIUM GAS FOR SYNTHESIS OF TRITIUM LABELLED COMPOUND

  • 摘要: <正> 氚标记化合物作为示踪剂,在医学、生物学、药理学、尤其是在生物工程研究中的广泛应用,已受到人们的重视。氚标记化合物的合成可以采用多种方法,微波激活法是其中一种重要的方法。微波激活法具有消耗氚气少,标记时间短,操作简便,标记产物杂质少,易纯化,较少破坏产物生物活性等一系列优点。近年来,这种标记技术发展很快。

     

    Abstract: An improved microwave tritium labelled apparatus was designed. It consists of a special vaccum circuit in which tritium gas can be cycled by a centrifugal mini-blower through the labelling reaction chamber. In this system a method of tritium labelling utilizing microwave activation of flowing tritium gas was developed. Using 8-BrcAMP as raw material, especially when 8-BrcAMP was precipitated on a SiO_2 carried Pd catalyst, the tritium-bromine replacement reaction occurred. The activity yield of ~3H-CAMP is more than that of ~3H-cAMP prepared by microwave tritium labelling without tritium gas circulating. It is an improved microwave method for tritium labelling on definite position of molecules.
